I haven’t given this any thought before but the manual tells you to set the idle speed with car in drive. Then set the mixture screws. I have always put the car in park or neutral when setting the mixture. But should I keep it in drive? How do you guys do it?

You should follow the manual. Leave it in Drive, (or reverse), set the idle speed, then adjust the mixture screws for the highest vacuum reading with the gauge tee'd into the distributor vac advance.

The mixture screws should be the LAST thing you adjust, after everything else has been adjusted and the engine is warm.
